Kambosos was born in Sydney, New South Wales, on June 14, 1993, to parents of Greek descent. His paternal grandparents immigrated to Australia from Sparta, Laconia, Greece, and Kambosos has the famous Spartan battle cry “Never retreat, never surrender” tattooed on his body in homage to his heritage. He started playing junior rugby league for Gymea Gorillas at a young age and was often ostracized as a child for being overweight. Hence, his father enrolled him in boxing classes at age 11 to improve his fitness. Kambosos quickly shed the excess weight and was placed in the Cronulla Sharks development squad, where Ricky Stewart coached him, but he found himself at a crossroads as a teenager, forced to choose between boxing and rugby league. He elected to follow the boxing path and began his fighting career with 85 wins in 100 amateur bouts before turning professional.

Kambosos made his professional boxing debut in May 2013 at 19. In December 2016, he defeated world number nine Brandon Ogilvie to win the WBA Oceania title. He then faced Kamil Balla in May 2017, whom he defeated by a unanimous decision in a ten-round fight. Five months later, he knocked out Cry Sethaphon in the ninth round and won the WBA Oceania and IBF Pan Pacific lightweight titles.
In April 2018, Kambosos signed a promotional deal with DiBella Entertainment.
In May 2018, he made his US debut. On January 19, 2019, Kambosos defeated Ray Perez at the MGM Grande in Las Vegas. On June 7 in Athens, Kambosos returned to his native Greece. After the win, he called former two-division world champion Jose Pedraza.
On December 14, 2019, Kambosos faced his toughest opponent, former IBF lightweight champion Mickey Bay.
On October 31, 2020, Kambosos defeated former IBF featherweight champion, Lee Selby by a split decision at the SSE Arena in London. At the time, Selby was ranked #1 by the WBO, #4 by the IBF, and #12 by the WBC. The win over Selby makes Kambosos a mandatory challenger for the IBF lightweight title.

On January 9, 2021, the IBF ordered undefeated unified lightweight champion Teofimo Lopez to defend his titles against Kambosos, their number one contender and mandatory challenger. The fight was originally scheduled for June 5, 2021, at LoanDepot Park in Miami, Florida, before being postponed multiple times due to complications with Lopez contracting COVID-19 and disputes over the fight venue. Kambosos won the bout via a split decision to become the new Unified Lightweight World Champion.
Kambosos vs Haney
On June 5, 2022, Kambosos Jr and Devin Haney clashed at Marvel Stadium in Melbourne, Australia, to determine the 1st Undisputed Lightweight Champion of the four-belt era. The fight did not proceed without controversy, as Kambosos could not make weight in his first attempt. He initially weighed in at 135.36 lbs, 0.36 lbs over the 135 lbs limit for the lightweight division. On his second attempt an hour later, within the two-hour limit, Kambosos weighed 134.49 lbs. The fight contract included an automatic rematch clause that Kambosos intends to exercise. A possible rematch would take place in late 2022 in Australia.
